• Rubicon assigns distance constraints from SMILES + SMARTS rules and generates initial random coordinates (distance geometry random coordinate method; Spellmeyer, 1997).
   
• Each conformer's center of mass is translated to a sphere chosen with probability proportional to its LIGSITE score.
